Study of chronic orofacial pain with preclinical models

چکیده
Background Chronic orofacial pain conditions such as trigeminal neuropathic pain, reflex sympathetic dystrophy of the face, and temporomandibular pain disorders are debilitating pain conditions. They are known to respond poorly to treatment including opioids in human patients. There is a pressing need to better understand the mechanisms of these chronic orofacial pain conditions so that new and effective therapeutic targets can be identify. To achieve this goal, preclinical animal models that well represent human chronic orofacial pain conditions are needed. In the present study we used two preclinical models of chronic orofacial pain, the chronic constriction nerve injury of the infraorbital nerve (ION-CCI model) and the oxaliplatin-induced orofacial pain (oxaliplatin model). We applied newly developed orofacial operant behavioral assessment to study orofacial pain and to test effects on Kv7.2 channel activators in alleviating orofacial pain in these animals.

Trigeminal Central Sensitization and Its Modulation in Acute and Chronic Orofacial Pain Models
This study aimed to examine whether trigeminal nerve injury induces chronic nociceptive behaviour and central sensitization (CS) in functionally identified medullary dorsal horn (MDH) nociceptive neurons in mice, and whether CS in acute and chronic orofacial pain models and nociceptive behaviour in the chronic model are affected by systemic administration of pregabalin. Infraorbital nerve injur...

Previous findings have shown that saffron (Crocus sativus L.) extract and its active constituents produce antinociceptive effects in the rat models of orofacial pain. In the present study, the central H2 histaminergic and alpha-2 adrenergic receptors involvement in crocetin-induced antinociception in orofacial formalin pain in rats was evaluated. The guide cannula was implant...
متن کاملIsobolographic antinociception of non-steroidal anti-inflammatory drugs in rodent formalin orofacial pain
Diverse studies suggest that non-steroidal anti-inflammatory drugs (NSAIDs) induce antinociception through inhibition of cyclooxygenases. In this study was evaluated the effect of NSAIDs to induce antinociception either alone or in combination in the formalin orofacial test of rodent. Male mice were injected intraperitoneally with dexibuprofen, dexktoprofen, diclofenac meloxicam, metamizol, and...

The year 2013-2014 has been designated the Global Year Against Orofacial Pain by the International Association for the Study of Pain. Accordingly, a multidisciplinary Canadian and international group of clinical, research and knowledge-transfer experts attended a workshop in Montreal, Quebec. The workshop had two aims: to identify new pathways for innovative diagnosis and management of chronic ...
متن کاملRole of opioid system in verapamil-induced antinociception in a rat model of orofacial pain
Calcium, through its various channels involves in local, spinal and supra-spinal transmission of pain. In the present study, we investigated the separate and combined treatment effects of verapamil (a calcium channel blocker), morphine (an opioid agonist) and naloxone (an opioid antagonist) on pain in the orofacial region of rats. Orofacial pain was induced by subcutaneous (SC) injection of for...
